CJC-1295 / Ipamorelin Benefits: What the Research Actually Shows

The one benefit of CJC-1295 with ipamorelin that human studies establish is a rise in your own growth hormone and IGF-1, with the pituitary's pulsing pattern kept intact. Sleep, recovery, body composition, bone and "anti-ageing" are inferred from what growth hormone does, supported by animal work and prescriber reports, and none has been measured in a trial of the pairing. There are no randomised controlled trials of the CJC-1295 and ipamorelin combination in humans at all. It is a compounded prescription injection at Pepti, and compounded medications are not FDA approved.

CJC-1295 / Ipamorelin Benefits
The short answer
| Claimed benefit | What the evidence is | Strength of evidence |
|---|---|---|
| Raises your own growth hormone | Human: 2006 CJC-1295 trials (DAC form) and the 1999 ipamorelin volunteer study | Established for each component |
| Raises IGF-1 | Human: 2006 trials reported IGF-1 up 1.5 to three-fold for nine to eleven days after one injection of the DAC form | Established for that form |
| Keeps the pituitary's pulses | Human: 2006 overnight-sampling study, pulses preserved with higher troughs | Moderate, one study |
| A bigger pulse than either alone | Human and mechanism: the two receptors are described as more than additive; a 1995 GHRH plus GH-releasing-peptide study | Moderate; the pairing itself untested |
| Better sleep | No trial; physiology plus prescriber reports | No direct evidence |
| Faster recovery | No trial; prescriber reports | No direct evidence |
| Leaner body composition | No trial of the pairing; one 2001 mouse study on ipamorelin ran the other way | No evidence, and some against |
| Bone | Animal: three rat papers on ipamorelin, 1999 to 2001 | Preclinical only |
| Skin, hair and general anti-ageing | No trial | No direct evidence |
| Fewer side effects than the older secretagogues | Animal: the 1998 pig study behind ipamorelin's selectivity | Preclinical, but a real design difference |
The honest framing, in one line: known to raise growth hormone and IGF-1 in humans, with everything beyond that inferred from physiology and clinical experience.
CJC-1295 / Ipamorelin Benefits
What is in the vial, and which CJC-1295 it is
| Component | What it is | Receptor |
|---|---|---|
| CJC-1295 | A growth-hormone-releasing hormone analogue: the GHRH (1-29) chain with four substitutions that protect it from the enzyme DPP-IV | GHRH receptor |
| Ipamorelin | A five-amino-acid synthetic peptide that imitates ghrelin | Growth hormone secretagogue receptor, GHS-R1a |
Neither contains growth hormone. Both ask your own pituitary to release what it already makes, with the body's brakes, somatostatin and IGF-1 feedback, still working. That is the central difference from injected growth hormone, explained in peptides vs HGH.
CJC-1295 exists in two forms, and this matters for reading the evidence. The version in the published human trials carries a drug affinity complex (DAC) that binds albumin and keeps it circulating for about a week. The version compounded in blends, including Pepti's, is CJC-1295 without DAC, also called modified GRF (1-29), which lasts roughly half an hour and is therefore dosed daily alongside ipamorelin for one clean pulse. So the human data below belongs to a longer-acting relative of what is actually in the vial. The difference is set out in CJC-1295 with DAC vs without DAC.
CJC-1295 / Ipamorelin Benefits
Benefit 1: More growth hormone, and more IGF-1
What the research describes. The central paper is a 2006 study in the Journal of Clinical Endocrinology and Metabolism: two randomised, placebo-controlled, double-blind ascending-dose studies in healthy adults aged 21 to 61, lasting 28 and 49 days, using the DAC form. A single subcutaneous injection raised mean growth hormone two- to ten-fold for six days or more and IGF-1 1.5- to three-fold for nine to eleven days. No serious adverse reactions were reported, and the authors described the drug as well tolerated, particularly at the lower two of the four doses. For ipamorelin, the 1999 Pharmaceutical Research study in healthy male volunteers reported dose-proportional kinetics, a half-life of about two hours and a single growth hormone peak at roughly 40 minutes.
Evidence type. Human.
What it means. This is the one thing shown in people, and it is a hormone measurement rather than an outcome. Growth hormone is short-lived in blood; most of what it does in adults it does by prompting the liver to make IGF-1, which acts on muscle, bone, connective tissue and fat. IGF-1 is also the marker a physician can measure, and the aim is a result inside the age-adjusted range rather than the highest attainable.
CJC-1295 / Ipamorelin Benefits
Benefit 2: The pulses are preserved
What the research describes. A second 2006 paper in the same journal sampled blood overnight in healthy men before and one week after a single injection of the DAC form. Pulse frequency and size did not change, but trough levels between pulses rose about 7.5-fold, mean growth hormone rose 46 per cent and IGF-1 rose 45 per cent.
Evidence type. Human, one study.
What it means. This is the basis for saying CJC-1295 preserves pulsatility, and it is the main reassurance against the fear that a long-acting GHRH analogue flattens the rhythm. A careful reader also notes that preserved pulses sat alongside a marked rise in the baseline between them, and that the study says nothing about months of sustained stimulation, which has not been examined. The no-DAC form prescribed today is designed to produce one short pulse per dose rather than a sustained signal.
CJC-1295 / Ipamorelin Benefits
Benefit 3: A bigger pulse than either peptide alone
What the research describes. The pituitary listens to two inputs on different receptors, and the classes have been described as more than additive since the early 1990s. A 1995 paper in Clinical Endocrinology reported a larger response from a GHRH plus GH-releasing-peptide combination than from either alone. Ipamorelin also reduces somatostatin at the hypothalamus, taking a brake off.
Evidence type. Human, but for the class rather than this pairing.
What it means. One dose of both, on an empty stomach, is intended to produce a single pulse resembling the body's own nocturnal one. No trial has measured the CJC-1295 and ipamorelin combination itself. Practically, moving from ipamorelin alone to the pairing changes the size of the signal, not just the number of peptides, which is why the dose signals in the safety section matter more rather than less.
CJC-1295 / Ipamorelin Benefits
Benefit 4: Sleep
What the research describes. Nothing directly. No published trial of this pairing has sleep as an endpoint.
Why it is claimed. The largest growth hormone pulse of the day occurs during slow-wave sleep, and a 2000 study in JAMA found slow-wave sleep and growth hormone secretion declining together in healthy men from early adulthood. Prescribers reason that restoring a nocturnal pulse may travel with deeper sleep, and sleep is the most common early report: falling asleep faster, waking less, sometimes within the first few nights, often with more vivid dreams.
Evidence type. None direct.
What it means. It is the most consistent thing patients describe and the least measured. Some people notice nothing at all, which is normal; growth hormone pulses are not something you feel. If sleep is the only goal, DSIP is aimed at it directly and the SERENITY Blend combines DSIP and Selank with this same pairing; peptides for sleep compares them.
CJC-1295 / Ipamorelin Benefits
Benefit 5: Recovery
What the research describes. Nothing directly for the pairing.
Evidence type. Prescriber reports, usually described from the second or third refill rather than the first, and dependent on sleep, protein and training, none of which the peptide replaces.
What it means. Growth hormone's effects on connective tissue are established from growth hormone itself, so this is an extrapolation along a plausible pathway. For a specific injury, secretagogues are not injury treatments: BPC-157 and TB-500 are what the recovery literature is about, and peptides for muscle growth and recovery sets out where each sits.
CJC-1295 / Ipamorelin Benefits
Benefit 6: Body composition
What is claimed. Fat loss and lean mass are the claims made most often for this pairing online.
What the research describes. No human trial of the pairing has measured either. Growth hormone's effects on lean mass, fat distribution and bone are established from growth hormone itself and from tesamorelin, a different GHRH analogue with randomised human trials on visceral fat. For the components here the direct work is in animals, and one piece of it runs against the claim: a 2001 paper in Biochemical and Biophysical Research Communications reported that in mice, secretagogues increased body weight, fat-pad weight, leptin and food intake, through mechanisms that likely include feeding.
Evidence type. No trial of the pairing; animal evidence pointing the other way for the ipamorelin half.
What it means. This is not an appetite medication and it is not a weight-loss medication; ipamorelin acts at the receptor ghrelin uses to signal hunger, so some people notice more appetite rather than less. If scale weight is the goal, semaglutide and tirzepatide have large randomised trials with weight as the primary endpoint; best GLP-1 for weight loss covers that. If visceral fat specifically is the target, tesamorelin benefits sets out the one growth-axis peptide with human trial data on it.
CJC-1295 / Ipamorelin Benefits
Benefit 7: Bone
What the research describes. Three rat papers on the ipamorelin half: increased longitudinal bone growth rate in adult female rats over 15 days (Growth Hormone & IGF Research, 1999), higher bone mineral content after 12 weeks attributed to larger bones rather than denser bone (Journal of Endocrinology, 2000), and protection against the loss of bone formation caused by three months of high-dose methylprednisolone (Growth Hormone & IGF Research, 2001). For CJC-1295, a 2006 paper in the American Journal of Physiology reported once-daily dosing normalising growth in mice engineered to lack GHRH entirely.
Evidence type. Animal.
Strength. No human counterpart. This is not a treatment for osteoporosis or for steroid-related bone loss.
CJC-1295 / Ipamorelin Benefits
Benefit 8: Skin, hair and "anti-ageing"
What the research describes. Nothing. No trial of this pairing has measured skin, hair, energy or any ageing endpoint, and nothing shows it extends life or healthspan in people. The label "anti-ageing" describes the category the product sits in, not a demonstrated effect.
What it means. If skin is the actual goal, the peptides with that literature behind them are elsewhere: GHK-Cu benefits covers the copper peptide, and REVIVE combines GHK-Cu and TB-500 with this same pairing in one vial.
CJC-1295 / Ipamorelin Benefits
Benefit 9: A cleaner profile than the older secretagogues
What the research describes. The 1998 founding paper on ipamorelin in the European Journal of Endocrinology tested this in pigs: GHRP-6 and GHRP-2 raised ACTH and cortisol, while ipamorelin did not, at doses more than 200-fold above the dose needed for half-maximal growth hormone release, and none of the compounds affected FSH, LH, prolactin or TSH. CJC-1295 acts on the GHRH receptor and is not associated with cortisol release.
Evidence type. Animal, for the selectivity claim.
What it means. It is a real design difference and the main reason this pairing rather than an older peptide is the common prescription, but in people the selectivity is inferred rather than measured. Hexarelin is the older relative that does raise cortisol and prolactin with each dose, and remains available where a physician wants that profile.

What the evidence does not establish
- That the pairing improves sleep, recovery or body composition. Those outcomes have not been measured in a trial of it.
- An effect size or a timeline. Anything said about results in a set number of weeks is prescriber experience.
- That the no-DAC form actually prescribed does what the DAC form did in 2006. Its human evidence is borrowed from the shared chain.
- Long-term safety with continuous use over years, including cancer incidence, which a 2018 class review in Sexual Medicine Reviews named as the specific gap.
- Whether the two together behave differently from the sum of the two apart.
- Efficacy for any condition as an approved treatment. Neither component is approved for anything.

Where it stands with the FDA
Both components were placed in Category 2 of the FDA's interim 503A bulk drug substances list in 2023 and left that category in September 2024 when their nominations were withdrawn and the FDA referred them to its Pharmacy Compounding Advisory Committee. On 29 October 2024 the committee voted 0 in favour and 12 against, with one abstention, on adding ipamorelin to the 503A Bulks List, for growth hormone deficiency and postoperative ileus. On 4 December 2024 it voted 0 to 13 against CJC-1295 (free base), 1 to 12 against the acetate and 0 to 13 against each of the three DAC forms, for growth hormone deficiency. The recorded reasoning was the absence of evidence for those uses, not a finding of harm. Those votes are advisory. Neither compound appears in any 503A category on the list updated 14 May 2026, no final determination has been issued, and both remain legal to prescribe and dispense. Compounded medications are not FDA approved. Are peptides FDA approved explains the difference.
Anti-doping
WADA's 2026 Prohibited List names CJC-1295 among GHRH analogues and ipamorelin among growth hormone secretagogues, under S2.2.4, prohibited at all times, in and out of competition. The list does not distinguish DAC from no-DAC, and a prescription does not create an exemption. See do peptides show up on a drug test.

Side effects, in brief
| What is reported | What it usually means | What to do |
|---|---|---|
| Injection-site redness, itching or a small lump | Common local reaction | Rotate sites |
| A brief flush or head-rush within minutes | A GHRH-receptor effect, most noticeable early | Report if marked |
| Headache early on | Reported in the 2006 trials and in practice | Report if persistent |
| Hunger after a dose | On-target: ipamorelin acts at the ghrelin receptor | Report it, especially if weight is a concern |
| Vivid dreams with evening dosing | Tracks the change in deep sleep | Report it |
| Fluid retention, puffy hands or ankles, joint aches, tingling hands | Growth hormone above what you need | Message your care team; do not change the dose yourself |
| Facial or throat swelling, widespread rash, breathing difficulty | A serious allergic reaction | Call 911 or get emergency care first, then tell your physician |
Because growth hormone opposes insulin, diabetes or impaired glucose tolerance is raised at intake, and because the IGF-1 axis supports tissue growth, a cancer history is raised. There is no established interaction list. With CJC-1295 in the picture a dose change shows more slowly than with ipamorelin alone, which is why a prescriber acts on the first report rather than waiting for a pattern. The typical directions for the single vial are 0.25 mL, which is 25 units on a U-100 insulin syringe, subcutaneously once daily Monday through Friday on an empty stomach. That is 0.3 mg of CJC-1295 and 0.6 mg of ipamorelin per dose, 20 doses per vial, about four weeks, which is why refills run every 28 days, one vial per fill. The empty-stomach instruction is not a formality: glucose and fatty acids after a meal blunt the pulse. Your physician sets your directions after reviewing your intake, and they are printed on the vial.
Every price is all-in, covering medication, physician review, refill management and shipping, with no separate membership fee. Pepti is cash-pay and does not bill insurance. Vials are multi-dose solutions shipped ready to use, refrigerated with ice packs, with sterile syringes and alcohol prep pads. Bloodwork is not required for any Pepti treatment. If your physician wants labs for your care, they'll ask through your account.
If you want the GHRH side shorter still, Ipamorelin + Sermorelin at $219 a month is the equivalent pairing built on sermorelin; CJC-1295 vs sermorelin and sermorelin vs tesamorelin vs ipamorelin compare the options, and PHYSIQ vs SHRED vs TITAN compares the blends.

Common questions
CJC-1295 / Ipamorelin Benefits, answered.
The established one is a rise in your own growth hormone and IGF-1, shown in human studies of each component. Sleep is the most common patient report, with recovery and body composition described later if at all. None of those has been measured in a trial of the pairing.
It does what human studies say it does: it raises growth hormone and IGF-1 through your own pituitary. Whether that translates into better sleep, recovery or body composition has not been tested in a trial, so that part rests on physiology and clinical experience.
They act on different receptors on the same pituitary cells, and the two classes are described as more than additive. Ipamorelin also takes a brake off at the hypothalamus. One dose of both is intended to produce a single fuller pulse.
No. It is not an appetite medication, and ipamorelin acts at the receptor ghrelin uses to signal hunger, so some people notice more appetite. For scale weight, the GLP-1 medications have the trial evidence.
No trial has measured muscle or strength. Prescribers describe recovery and body-composition changes where training, protein and sleep are in place, which is a report rather than a finding.
It is the most common early report, and the physiology is plausible because the largest natural growth hormone pulse arrives in deep sleep. No published trial of the pairing has sleep as an endpoint.
There is no trial-established timeline. Prescribers describe sleep reports in the first weeks and anything else over the second or third refill. Response varies widely, and anyone quoting specific numbers is selling rather than prescribing.
Not for the combination. The human evidence is the 2006 healthy-adult trials of the DAC form of CJC-1295, the 1999 ipamorelin volunteer study and the 2014 postoperative ileus trial. None measured sleep, recovery or body composition.
No. Both components prompt your pituitary to release its own growth hormone and the feedback loop through somatostatin stays intact; the 2006 overnight study reported pulses preserved. That is the opposite of injected growth hormone, which suppresses the pituitary's own output while it is present.
The version without DAC, also called modified GRF (1-29), 6 mg per 5 mL vial alongside 12 mg of ipamorelin. The published human trials used the DAC form. Your label states what you are prescribed.
No. Compounded medications are not FDA approved, and neither component has ever been an approved drug. The FDA's advisory committee voted against adding both to the 503A Bulks List in late 2024 on grounds of insufficient evidence, and no final determination has been issued.
At Pepti, $239 a month for the vial, all-in, covering medication, physician review, refill management and shipping. The blends containing the pairing run $279 to $289.
