AOD-9604 vs 5-Amino-1MQ: Mechanism, Evidence, FDA Status, Cost, and Which One Fits
This page explains each on its own terms, puts them side by side, covers what the research does and does not show, where each stands with the FDA, how each is dosed, what each costs at Pepti, and who tends to suit which.

Treatments mentioned
AOD-9604 and 5-Amino-1MQ are both prescribed around fat metabolism, but they are different kinds of molecule acting on different targets, with different gaps in their evidence. AOD-9604 is a 16-amino-acid fragment of growth hormone described around fat-cell signalling in mice; it went through six human trials, and the largest found no significant weight loss over placebo. 5-Amino-1MQ is not a peptide but a small synthetic molecule that blocks the fat-cell enzyme NNMT; it has never been tested in a human trial of any kind. Neither acts on appetite, both are prescription-only and compounded by a licensed US pharmacy, and neither is FDA approved.

AOD-9604 vs 5-Amino-1MQ
The short answer
| If your main goal is | The usual prescription | What is in it | Price |
|---|---|---|---|
| A fat-metabolism signal with some human safety data | AOD-9604 | 10 mg AOD-9604 in a 5 mL vial | $229/mo |
| Fat-cell metabolism through the NNMT enzyme and the NAD+ angle | 5-Amino-1MQ | 5 mg 5-Amino-1MQ in a 5 mL vial | $219/mo |
| AOD-9604 with growth hormone secretagogues and MOTS-c | SHRED Blend | AOD-9604, CJC-1295, ipamorelin and MOTS-c | $279/mo |
| 5-Amino-1MQ with BPC-157, MOTS-c and tesamorelin | FUSION Blend | BPC-157, MOTS-c, 5-Amino-1MQ and tesamorelin | $279/mo |
| Appetite and body weight, with human trial evidence | Semaglutide or tirzepatide | Dose-specific vials | From $99/mo or from $159/mo |
Every figure is an all-in monthly price covering the medication, physician review, refill management and shipping. There is no separate membership fee. GLP-1 prices are "from" prices because the vials are dose-specific. Current pricing always lives on each product page.
AOD-9604 vs 5-Amino-1MQ
AOD-9604 on its own terms
What it is
Human growth hormone is a protein of 191 amino acids. AOD-9604 is a synthetic copy of the last fifteen, positions 177 to 191, with a tyrosine added at the front, making a 16-amino-acid peptide held together by one disulphide bridge. It is the molecule sold online as "hGH fragment 176-191". AOD stands for Anti-Obesity Drug; it came out of Monash University in Melbourne and was developed in the late 1990s by Metabolic Pharmaceuticals as an oral obesity treatment. It is not growth hormone and not a secretagogue: cell work reports it does not bind the growth hormone receptor, and IGF-1 did not change in the human trials. The full reference is at what is AOD-9604.
How AOD-9604 is described to work
- The "lipolytic domain" idea. A 2000 paper in Hormone Research proposed that growth hormone's effect on fat lives in its tail end, separable from its growth and blood-sugar effects.
- Beta-3 adrenergic receptors. A 2001 paper in Endocrinology reported reduced weight gain and more beta-3 receptors in the fat of obese mice, with no effect in mice lacking that receptor.
- Fat oxidation without a glucose rise. A 2001 paper in the International Journal of Obesity reported increased fat oxidation and plasma glycerol in obese mice, without growth hormone's rise in blood glucose.
The FDA's 2024 review noted that the molecular target has never been identified and that the animal studies did not establish a dose-response relationship.
What human data exists for AOD-9604
Six randomised, placebo-controlled trials, run in Australia between roughly 2001 and 2007, enrolling close to 900 people. The largest, OPTIONS, gave 536 adults with obesity oral AOD-9604 or placebo daily for 24 weeks alongside a supervised diet and exercise program and did not find statistically significant weight loss versus placebo; the company ended development for obesity in February 2007. What the trials did establish is mostly safety: no change in IGF-1, no impairment of glucose tolerance, no antibodies detected in the subjects tested, and no serious adverse event judged related to treatment. Every trial was oral or intravenous, not the subcutaneous injection that is prescribed.
AOD-9604 vs 5-Amino-1MQ
5-Amino-1MQ on its own terms
What it is
5-Amino-1MQ is short for 5-amino-1-methylquinolinium. It is a small synthetic molecule, not a peptide: there is no chain of amino acids in it, just a two-ring quinolinium core with a methyl group and an amino group attached. It came from a medicinal chemistry programme at the University of Texas Medical Branch in Galveston, which screened compounds against the enzyme nicotinamide N-methyltransferase (NNMT) in a 2017 Journal of Medicinal Chemistry paper. The reason anyone looked was a 2014 paper in Nature reporting that knocking NNMT down protected mice against diet-induced obesity. The full reference is at what is 5-Amino-1MQ, and the chemistry question is answered in is 5-Amino-1MQ a peptide.
How 5-Amino-1MQ is described to work
- Blocking NNMT. NNMT takes nicotinamide, the form of vitamin B3 cells recycle into NAD+, and attaches a methyl group from SAM, so the product is excreted rather than reused. Blocking it is proposed to keep both the NAD+ precursor and SAM in the cell.
- NAD+ and SAM in cultured fat cells. A 2018 paper in Biochemical Pharmacology reported higher intracellular NAD+ and SAM and less fat-building in cultured adipocytes. This is the origin of the "raises NAD+" description, and it is a cell-culture result.
- Weight and fat in obese mice. The same 2018 work reported lower body weight and white fat mass in diet-induced obese mice with food intake unchanged. A 2024 paper in Diabetes, Obesity and Metabolism dosed it once daily for 28 days and reported limited fat gain and improved glucose tolerance in obese mice, plus distribution to fat, muscle and liver after subcutaneous injection.
What human data exists for 5-Amino-1MQ
None. There are no human trials of any design, and as of September 2026 none registered under its name. The only human data is about the target, not the drug: a 2015 Diabetologia study reported roughly twofold higher NNMT expression in the fat tissue of people with type 2 diabetes. That is a reason to study the compound in people, not evidence that it works in them.

The real difference is what kind of unknown you accept
People ask which is "stronger" for fat. No study has measured both, and neither has a positive human result for fat loss, so that question has no answer. The useful distinction is the kind of uncertainty. AOD-9604 has been tested in people and the answer for weight was no, though the same trials gave it a reasonably clean safety record by mouth and by vein. 5-Amino-1MQ has never been tested in people at all, so the question is simply open, with no human safety record either. One has a known negative; the other has a blank.
AOD-9604 vs 5-Amino-1MQ
Who suits which
AOD-9604
AOD-9604 suits someone who wants a fat-metabolism signal that leaves appetite and the growth hormone axis alone, who is already training and eating well, and who values having some human safety data even though the efficacy trials were negative. Prescribers typically describe it as support for a program that is already working. It is also part of the SHRED and PHYSIQ blends, which pair it with growth hormone secretagogues.
5-Amino-1MQ
5-Amino-1MQ suits someone whose interest is fat-cell metabolism or the NAD+ angle, with diet and activity already in place, and who is comfortable with a compound that has only mouse and cell evidence. Both of its main mouse studies paired it with a lifestyle change and reported the combination doing more than either alone. Its dose per injection is half that of AOD-9604, because the vial is half the concentration.
Both together
They act on different targets, and some physicians combine them. No Pepti product contains both: AOD-9604 is in SHRED and PHYSIQ, 5-Amino-1MQ is in FUSION. No study has tested the combination, so it means two unknowns at once. Most people start with one and add the second only if there is a reason to. The general rules are in can you take two peptides together.
AOD-9604 vs 5-Amino-1MQ
When neither is the right first step
| Situation | What makes more sense |
|---|---|
| Losing a meaningful amount of weight is the goal | Semaglutide or tirzepatide, which have large randomised human trials with weight as the endpoint |
| Fat around the middle specifically | Tesamorelin, with human trial data on visceral fat; see AOD-9604 vs tesamorelin |
| NAD+ itself is the interest | NAD+ injection, which supplies the molecule directly |
| Muscle fuel use and the exercise signal | MOTS-c or SLU-PP-332; see MOTS-c vs SLU-PP-332 vs 5-Amino-1MQ |
| A fat-transport cofactor with human trials | L-Carnitine Injection, whose oral trials have been pooled in meta-analyses |

Where each stands with the FDA
The two are in different places, and most pages online have not caught up.
| AOD-9604 | 5-Amino-1MQ | |
|---|---|---|
| 2023 | Placed in Category 2 of the 503A interim list | Not on any list |
| 2024 | Left Category 2 on 27 September 2024 when nominations were withdrawn; advisory committee voted 0 to 12 against on 4 December 2024, for obesity | Not on any list |
| Interim list updated 14 May 2026 | Not in Category 1, 2 or 3; listed as "nominated but withdrawn" | Not in Category 1, 2 or 3 |
| July 2026 advisory committee | Not on the agenda | Not on the agenda |
| Final FDA determination | None issued | Never evaluated |
A committee vote is a recommendation, not a decision. AOD-9604 has been looked at and received a negative recommendation; 5-Amino-1MQ has simply never been through the FDA's evaluation. Both remain legal to prescribe and dispense as compounded medications, and compounded medications are not FDA approved. Are peptides FDA approved explains the difference between not approved and not legal.
AOD-9604 vs 5-Amino-1MQ
Dosing, side by side
Both are injected under the skin (subcutaneously) into the abdomen or thigh, rotating sites, once a day Monday to Friday with weekends off. The volume is the same, 0.25 mL, which is 25 units on a U-100 insulin syringe. Because the vials differ in strength, that volume delivers 0.5 mg of AOD-9604 or 0.25 mg of 5-Amino-1MQ. Some prescribers direct AOD-9604 in the morning before eating. Twenty doses per vial covers about four weeks, which is why refills run every 28 days, one vial per fill.
You do not need to calculate anything: your physician sets your directions after reviewing your intake, and your directions are printed on the vial. Both ship refrigerated with ice packs, with sterile syringes and alcohol prep pads. Bloodwork is not required for any Pepti treatment. If your physician wants labs for your care, they'll ask through your account. The conversions are in AOD-9604 dosage and 5-Amino-1MQ dosage.
AOD-9604 vs 5-Amino-1MQ
Side effects
AOD-9604. The six company trials reported headache, fatigue, dizziness and low blood sugar with intravenous dosing, and headache, diarrhoea, flatulence, increased appetite and nausea with oral dosing, with adverse events similarly distributed between peptide and placebo. With the injection, prescribers mostly report injection-site redness, mild soreness or bruising. No human trial has used the subcutaneous route.
5-Amino-1MQ. There is no human safety database. The mouse studies reported no observable adverse effects at the doses used, though they were not toxicology studies. Clinically, mostly injection-site reactions. NNMT also methylates some foreign compounds, so blocking it could in principle change how some drugs are handled; nothing is known about whether that matters, which is a reason to list every medication and supplement you take.
More detail is in AOD-9604 side effects and 5-Amino-1MQ side effects. For either, stop and contact your physician for any reaction that is severe, spreading or involves difficulty breathing. For swelling of the face or throat or trouble breathing, call 911.
AOD-9604 vs 5-Amino-1MQ
Who should not take either, or should discuss it first
| Situation | Why |
|---|---|
| Pregnancy or breastfeeding | Not used; safety data is absent for both |
| Tested athletes | AOD-9604 is prohibited by WADA as a growth hormone fragment; check the current list for 5-Amino-1MQ |
| Taking several prescription medications | Especially for 5-Amino-1MQ, because of the theoretical NNMT interaction point |
| On glucose-lowering medication | Raise it at intake; low blood sugar was reported with intravenous AOD-9604 |
AOD-9604 vs 5-Amino-1MQ
What the evidence shows, honestly
AOD-9604 has the larger human record of the two: six randomised trials, a clean safety profile by mouth and by vein, and a negative result for weight loss, with nothing for the subcutaneous route. 5-Amino-1MQ has mouse and cell studies from essentially one research group and human data only on its target enzyme. Neither has been shown to change weight or body fat in people. Anyone describing either as a trial-backed fat-burner is overstating what exists, and the appropriate frame is an adjunct to diet and training, not a replacement for either. The wider picture is in best peptides for weight loss.

Where to get AOD-9604 or 5-Amino-1MQ prescribed
Neither can be bought legitimately without a prescription. Sites selling them "for research use only" are not accountable for what is in the vial: identity, purity, sterility and concentration are untested. The prescription route:
- Complete a free online intake covering your history, medications and what you want help with.
- A physician licensed in your state reviews it, usually within 24 hours.
- If it is appropriate, a PCAB-accredited, FDA-registered US 503A pharmacy compounds it to that prescription. Lot test results are published at /quality/lab-results.
- It ships refrigerated with your directions printed on the vial.
- Your physician stays reachable for questions and dose changes.
Pepti prescribes both in every state except Oklahoma, plus DC, cash-pay, without billing insurance. Step-by-step pages: how to get AOD-9604 prescribed online and how to get 5-Amino-1MQ prescribed online.

Common questions
AOD-9604 vs 5-Amino-1MQ, answered.
Neither has a positive human result for fat loss. AOD-9604's largest human trial found no significant weight loss over placebo, and 5-Amino-1MQ has never been tested in people. No study has compared them.
No. AOD-9604 is a 16-amino-acid peptide. 5-Amino-1MQ is a small synthetic molecule with no amino acids in it, grouped with peptides because of how it is prescribed and injected.
Some physicians combine them because they act on different targets, but no study has tested the combination and no Pepti product contains both. Starting with one makes it easier to tell what is doing what.
No. Neither is described as acting on appetite, and the 5-Amino-1MQ mouse work specifically reported food intake unchanged.
AOD-9604 has more human safety data, from close to 900 trial participants on oral or intravenous dosing, with adverse events similar to placebo. 5-Amino-1MQ has no human safety data at all. Neither has been studied by subcutaneous injection in people.
5-Amino-1MQ is the one described around NAD+: NNMT inhibitors raised NAD+ inside cultured fat cells. That has not been measured in people. AOD-9604 has no described NAD+ effect.
AOD-9604 is in the SHRED and PHYSIQ Blends. 5-Amino-1MQ is in the FUSION Blend. No blend contains both.
Yes, with a valid prescription, as compounded medications from a licensed US pharmacy. Neither is FDA approved. AOD-9604 received a negative advisory committee recommendation in 2024 with no final rule; 5-Amino-1MQ has never been evaluated.
