Semax vs DSIP: Daytime Focus Peptide vs Bedtime Sleep Peptide, Compared

This page explains each peptide on its own terms, puts them side by side, covers what the research does and does not show, where each stands with the FDA after the July 2026 meeting, how each is dosed at Pepti, what each costs, and how to tell which question you are actually asking.

DSIP (Delta Sleep Inducing Peptide) (Injectable)
DSIP (Delta Sleep Inducing Peptide)$249/mo

Semax and DSIP are not alternatives for the same problem: Semax is a daytime peptide prescribed around focus and mental energy, and DSIP is a bedtime peptide prescribed around sleep. Semax is a seven-amino-acid fragment of ACTH, described around BDNF in the brain, with decades of use as a registered medicine in Russia. DSIP (delta sleep-inducing peptide) is a nine-amino-acid peptide first found in sleeping rabbits, described around deep-sleep EEG patterns and the stress response, with a handful of small 1980s intravenous trials in people with insomnia that gave mixed results. In July 2026 the FDA's advisory committee voted in favour of Semax and against DSIP. Both are prescription-only, compounded by a licensed US pharmacy, and neither is FDA approved.

Semax vs DSIP

Semax on its own terms

  • What it is

Semax is a heptapeptide: Met-Glu-His-Phe-Pro-Gly-Pro. The first four amino acids are positions 4 to 7 of adrenocorticotropic hormone (ACTH), the pituitary hormone that signals the adrenal glands to make cortisol. The last three, Pro-Gly-Pro, were added to slow its breakdown. The design keeps the part of ACTH studied for attention and learning and drops the part that raises cortisol, so Semax is described as having no corticotropic activity. It is not a steroid and not a stimulant. The full reference is what is Semax.

It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and is a registered medicine in Russia, where it has been used in neurology since the 1990s for acute ischaemic stroke and chronic cerebrovascular disease.

  • How Semax is described to work

    • BDNF and TrkB. A 2006 paper in Brain Research reported that Semax raised BDNF protein and activated its receptor, TrkB, in the rat hippocampus. BDNF is the growth factor most closely tied to synaptic plasticity, which is the basis of the "cognitive" label.
    • Neurotrophins in injured tissue. A 2010 paper in Cellular and Molecular Neurobiology reported increased transcription of BDNF, NGF and their receptors in the ischaemic rat cortex.
    • Serotonin and dopamine signalling. Animal work describes increased serotonergic signalling, and a mouse study reported Semax increasing amphetamine-induced dopamine release.

All of the mechanism work is in animals.

  • What human data exists for Semax

The human work is Russian clinical research, mostly in stroke and cerebrovascular patients, using the nasal medicine sold there: a 1997 study of 30 acute stroke patients against 80 matched controls, a 2005 study of 187 patients with chronic cerebrovascular disease that reported good tolerability, a 2018 trial in 110 stroke-rehabilitation patients in which plasma BDNF rose, and a 2018 single-dose functional MRI study in 14 healthy adults. There is no randomized, placebo-controlled trial in an English-language peer-reviewed journal, and no trial of Semax for focus or memory in healthy adults.

Semax vs DSIP

DSIP on its own terms

  • What it is

DSIP is a nonapeptide, nine amino acids long: Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu. In the early 1960s Marcel Monnier's group in Basel reported that blood from the brain of a sleeping rabbit could induce sleep-like EEG patterns in a second rabbit. The factor was isolated and synthesised by 1977, and a Proceedings of the National Academy of Sciences paper that year named it. The World Health Organization later gave it the nonproprietary name emideltide, which is the name the FDA uses. It is not a hormone, not related to melatonin, and not a sedative. The full reference is what is DSIP.

  • How DSIP is described to work

    • Deep-sleep EEG patterns. The defining observation, repeated in rabbits, rats and cats from 1977, is more delta-wave and spindle activity, the EEG hallmarks of deep non-REM sleep. It was not universal; one 1980 rat study found no change.
    • An indirect opioid route. In rats the opioid blocker naloxone stopped DSIP prolonging sleep. Cell work found DSIP does not bind opioid receptors directly but was reported to trigger release of the body's own enkephalins.
    • The stress axis. A 1985 paper in Neuroendocrinology reported DSIP reduced stress-hormone release in rats, and a 2001 paper in Stress found reduced stress-induced activation in the rat hypothalamus.
    • A very short life. DSIP lasts about eight minutes in human plasma before blood enzymes break it apart. Any effect is a signalling event, not a level held through the night.

Nearly fifty years on, no gene encoding DSIP and no DSIP receptor has been identified, and a 2006 review in the Journal of Neurochemistry called the sleep-factor hypothesis "extremely poorly documented and still weak."

  • What human data exists for DSIP

DSIP is unusual among compounded peptides because it has small randomized, double-blind, placebo-controlled studies in people, all intravenous and all from 1981 to 1992. The first, in the Lancet in 1981, gave six chronic insomniacs an infusion or placebo and reported longer, less interrupted sleep without daytime sedation. Two independent groups did not agree: a 1987 crossover study in the International Journal of Clinical Pharmacology Research found no significant difference from placebo nights, and a 1992 study of sixteen insomniacs in Neuropsychobiology judged the effects weak. The FDA counted roughly 209 people across all of this work. There are no trials of the subcutaneous route.

They answer different questions

Both get listed as brain peptides, but in practice they sit at opposite ends of the day. Semax is described around alertness, and some people feel over-alert if they take it late, which is why morning dosing is the usual direction. DSIP is described around the structure of sleep and is taken at bedtime. Which is "better" depends entirely on which part of the day is the problem.

Semax vs DSIP

Who suits which

  • Semax

Semax is the usual conversation when the complaint is daytime: difficulty concentrating, mental fatigue in the afternoon, trouble getting started on tasks, while sleep itself is reasonable. It has the longer record of human use and a clearer mechanism story. Semax benefits sets out what the research has looked at, and Selank vs Semax covers its calmer sibling.

  • DSIP

DSIP is the usual conversation when nights are the problem: waking repeatedly, sleep that feels light, or poor sleep layered on top of a stressful stretch. The animal work on the stress axis leans toward it being most relevant when stress is driving the poor sleep; long-standing chronic insomnia is the population where the human studies split. DSIP benefits covers the research in more detail.

  • When poor focus is really poor sleep

Brain fog after bad nights is a sleep question first. The Semax reference itself lists poor sleep as the case where DSIP or the SERENITY Blend is the conversation to have instead. Be honest on your intake about how you sleep.

  • Can you take Semax and DSIP together?

They are taken at different times of day and act through different described pathways. No study has tested the pairing, so it means two unknowns at once. Whether a combination fits you is something your physician sets after reviewing your intake, and starting with one makes it easier to tell what is doing what.

Semax vs DSIP

When neither is the right first step

Situation What makes more sense
You have not tried CBT-I for insomnia Cognitive behavioural therapy for insomnia is the first-line recommendation of every major sleep guideline
Loud snoring or pauses in breathing at night A sleep apnoea assessment; DSIP is not a treatment for breathing-related sleep problems
Anxiety keeps you awake or distracted Selank, described around anxiety signalling
Your sleep timing is off rather than its depth Epithalon, described around the pineal gland and circadian timing
New or worsening memory problems A medical work-up first; thyroid, B12, sleep, mood and medications can all cause it
ADHD Your prescribing physician; there is no evidence Semax works for ADHD

Semax vs DSIP

Where each stands with the FDA

Both were placed in Category 2 of the FDA's 503A bulk substances list in 2023, both were moved out of Category 2 in April 2026, and both were reviewed at the same Pharmacy Compounding Advisory Committee meeting on 24 July 2026. The votes went in opposite directions.

Semax DSIP (emideltide)
2023 Category 2 Category 2
April 2026 Moved out of Category 2 Removed from Category 2 on 15 April 2026
FDA staff proposal Not to add to the 503A bulks list Not to add to the 503A bulks list
24 July 2026 advisory vote 8 to 5, one abstention, recommended adding it 6 to 7, one abstention, did not recommend adding it
Final FDA determination Pending Pending

An advisory vote is a recommendation, not a decision. Neither peptide is banned, and both remain legal to prescribe and dispense as compounded medications. DSIP was the only one of the seven peptides considered that week not to receive a favourable vote, so unlike Semax it has no current path toward the positive list unless the FDA departs from the committee's advice. Neither is an FDA-approved drug. Are peptides FDA approved explains the difference between not approved and not legal.

Semax vs DSIP

Dosing, side by side

Both are injected under the skin (subcutaneously) into the abdomen or thigh, rotating sites, five days a week with weekends off. The draw is the same, 0.25 mL, which is 25 units on a U-100 insulin syringe. Because the vials differ in strength, that draw delivers different amounts. This is math only, for reading a label:

Semax DSIP
Directions printed on the vial Inject 0.25 mL (25 units) subcutaneously once daily Monday through Friday Inject 0.25 mL (25 units) subcutaneously Monday through Friday nights at bedtime
Concentration 2 mg/mL 1 mg/mL
Amount in 25 units 0.5 mg 0.25 mg
Total in the vial 10 mg in 5 mL 5 mg in 5 mL
Doses per vial 20 (5 mL divided by 0.25 mL) 20 (5 mL divided by 0.25 mL)
Time per vial 4 weeks at 5 doses a week 4 weeks at 5 doses a week

You do not need to calculate anything: your physician sets your directions after reviewing your intake, and they are printed on the vial. Never carry a units number from one product to another, because the same 25 units holds twice as much Semax as DSIP. Twenty doses covers about four weeks, which is why refills run every 28 days, one vial per fill. Both ship refrigerated with ice packs, sterile syringes and alcohol prep pads, as ready-to-use multi-dose solutions. Full breakdowns are in Semax dosage and DSIP dosage.

Semax vs DSIP

Side effects

Semax. The Russian clinical series describe it as well tolerated, including in older stroke patients. Reported with the injection: redness, mild soreness or bruising at the injection site, occasional early headache, and restlessness if it is taken late in the day. Anyone taking a stimulant should say so on the intake because of the mouse dopamine finding. More in Semax side effects.

DSIP. In the small intravenous insomnia studies it was reported as well tolerated. The withdrawal series reported transient headache, nausea and vertigo in a handful of patients. What prescribers report today is mostly injection-site redness or soreness, and sometimes an early headache or vivid dreams. Opioids, benzodiazepines, other sedatives and regular alcohol use all belong on the intake, because the mechanism is described through the body's own opioid system, and laboratory work reports DSIP being broken down by an enzyme some blood-pressure medicines inhibit. More in DSIP side effects.

For either, message your care team about anything that persists or worries you. Stop and call 911 for facial or throat swelling, widespread hives or difficulty breathing.

Semax vs DSIP

Who should not take either, or should discuss it first

Situation Why
Pregnancy or breastfeeding Not used; safety data is absent
Taking a stimulant Especially for Semax, because of the mouse dopamine finding
Taking opioids, benzodiazepines or other sedatives Especially for DSIP, because of its described opioid-system route
Taking blood-pressure medicines Especially for DSIP, because of the enzyme finding
Tested athletes Check your sport's current prohibited list before starting

Semax vs DSIP

What the evidence shows, honestly

Neither peptide has been shown in a Western-standard trial to do what it is prescribed for. Semax has a coherent BDNF mechanism, decades of human use as a registered medicine in Russia, and clinical studies in brain injury, but nothing in healthy adults for focus. DSIP has something rarer for a compounded peptide, small placebo-controlled human studies, but they are old, intravenous, tiny, and split between a positive originating group and two independent groups that found the effect too weak to matter. The controlled studies that measured slow-wave sleep did not find DSIP increased it in people. Any provider claiming either peptide has been shown to work is overstating the record.

Semax vs DSIP

Where to get Semax or DSIP prescribed

Neither can be bought legitimately without a prescription. Sites selling them "for research use only" are not accountable for what is in the vial: identity, purity, sterility and concentration are untested. The prescription route:

  1. Complete a free online intake covering your history, medications, sleep and what you want help with.
  2. A physician licensed in your state reviews it, usually within 24 hours.
  3. If it is appropriate, a PCAB-accredited, FDA-registered US 503A compounding pharmacy prepares it to that prescription. Lot test results are published at /quality/lab-results.
  4. It ships refrigerated with your directions printed on the vial.
  5. Your physician stays reachable for questions and dose changes.

Common questions

Semax vs DSIP, answered.

DSIP is the one prescribed around sleep and is taken at bedtime. Semax is a daytime focus peptide, and some people feel restless if they take it late. For sleep, DSIP or the SERENITY Blend is the usual conversation, though DSIP's human sleep studies were small and mixed.
