The science

How IGF-1 LR3 works, and what the research shows.

How it works

IGF-1 LR3 is a modified insulin-like growth factor 1 with an arginine substitution and 13-amino-acid extension that sharply reduce binding to IGF-binding proteins — keeping more of the peptide free and active, with a substantially longer effective half-life than native IGF-1.

It signals through the IGF-1 receptor, the body's primary mediator of growth-hormone-driven tissue growth.

Clinical evidence

  • Native IGF-1 (mecasermin) is FDA-approved for severe primary IGF-1 deficiency, which anchors the receptor biology in approved human medicine.

  • The LR3 analog itself, however, has no human clinical trials — its anabolic data comes from laboratory and animal research, making it one of the more investigational peptides physicians prescribe.
