How Long Does SYNAPSE Blend Take to Work? What Each Component's Studies Measured, and When

No one can tell you how long SYNAPSE takes to work, because the four-peptide blend has never been studied, in people or in animals. Its components' studies ran on very different clocks: single-dose Semax brain-imaging and rat Selank gene studies, Russian Selank anxiety trials over courses of weeks, MOTS-c work in mice only, and tesamorelin trials that measured cognition at 10 and 20 weeks and visceral fat on CT at 26 weeks, at doses two to four times the tesamorelin in a SYNAPSE dose. Those are study endpoints in named populations, not a promise of when anything will change for you. SYNAPSE is a compounded prescription, and compounded medications are not FDA approved.

Why there is no SYNAPSE timeline
The blend has never been studied
SYNAPSE is four peptides dissolved together in one vial: 5 mg Semax, 5 mg Selank, 10 mg MOTS-c and 10 mg tesamorelin. The name is a brand name for the combination and does not appear in any journal. Nobody has given the four together and measured anything, so "how long does SYNAPSE take" breaks into four questions, and even those do not add up to an answer, because no study has compared the blend with its components. Component by component
Semax: single doses and stroke courses
The studies that measured something soon after a dose measured a brain marker, not an experience: a 2006 Brain Research paper reported more BDNF protein and TrkB receptor activation in the rat hippocampus, and the 2018 study of 14 healthy adults reported changes on functional MRI after one intranasal dose. The studies that ran for weeks enrolled people recovering from a stroke or living with cerebrovascular disease. Selank: courses of weeks, in anxiety disorders
Selank's human studies are three small Russian comparator trials of 60 to 70 patients each, which measured anxiety over treatment courses of weeks against a benzodiazepine, not placebo; one reported the effect persisting about a week after the last dose. The fast timings, such as gene changes an hour after a dose, come from rats. None of the human work used injection. More in how long does Selank take to work.
MOTS-c: no human clock at all
There are no randomised controlled trials of MOTS-c in people. The treatment evidence is in mice, and the human data is observational: a bout of exercise raised the body's own MOTS-c in muscle and blood. There is nothing to time. More in how long does MOTS-c take to work.
Tesamorelin: the only component measured in months
Tesamorelin has the most specific timing evidence. IGF-1, the blood marker of the growth hormone signal, rose 81% over 26 weeks in the 2007 trial and 117% over 20 weeks in the cognition trial. The cognition trial looked at weeks 10 and 20 and reported its favourable effect on a composite at the end. The visceral fat trials measured at 26 weeks, with an extension to 52 weeks in which the reduction was maintained on treatment and returned toward baseline in those switched to placebo. Tesamorelin's pooled extension is the one data point on stopping: visceral fat returned toward baseline in people switched to placebo. What the evidence shows, honestly
The accurate framing for SYNAPSE is "one component with human trials for other indications, two with foreign clinical data in patient populations, one with animal data only," and no study of the four together. That means no effect size and no timeline in humans. Timelines given by prescribers are clinical experience, not trial endpoints, and no one can promise when, or whether, you will notice anything.
Where the components stand with the FDA: tesamorelin is the active ingredient of an FDA-approved product, but compounded tesamorelin is not that product; in July 2026 the Pharmacy Compounding Advisory Committee voted to recommend adding Semax and MOTS-c to the 503A Bulks List, with the FDA's final determinations pending; Selank's nomination was withdrawn and it has never been reviewed.
