5-Amino-1MQ Benefits: What the Research Actually Shows
This page takes each claimed benefit in turn and says what was studied, in what, and how much weight it can bear. It is organised by benefit, not by molecule; for the chemistry, dosing and the FDA position in full, read what is 5-Amino-1MQ.

Treatments mentioned
The benefits claimed for 5-Amino-1MQ, less fat gain, higher NAD+, better blood sugar handling, less fatty liver and stronger aged muscle, all come from cultured cells and mice. There are no human trials of 5-Amino-1MQ of any design. The only human data concerns its target, not the compound: a 2015 study reported higher levels of the enzyme NNMT in the fat tissue of people with type 2 diabetes. 5-Amino-1MQ is a small molecule, not a peptide, supplied at Pepti as a compounded prescription injection; compounded medications are not FDA approved.

What 5-Amino-1MQ is, in one paragraph
5-Amino-1MQ is short for 5-amino-1-methylquinolinium, a small synthetic molecule built on a quinolinium ring, with no amino acids in it; it is not a peptide (is 5-Amino-1MQ a peptide explains why). It came from a medicinal chemistry program at the University of Texas Medical Branch in Galveston, led by Stanley Watowich. It blocks the enzyme nicotinamide N-methyltransferase (NNMT). NNMT takes nicotinamide, the form of vitamin B3 cells recycle into NAD+, and attaches a methyl group from S-adenosylmethionine (SAM), the cell's main methyl donor; the product is excreted rather than reused. Blocking the enzyme is proposed to keep both the NAD+ precursor and SAM in the cell. It is not a hormone, not a GLP-1 medication, not a stimulant, and does not contain NAD+. Every benefit below hangs off that one enzyme.

Why anyone went looking
The idea came from a 2014 paper in Nature from Barbara Kahn's laboratory at Beth Israel Deaconess and Harvard, which reported NNMT expression rising in the white fat and liver of obese and diabetic mice, and reported that knocking the enzyme down protected mice against diet-induced obesity. Knocking it down was reported to increase energy expenditure in fat cells, with SAM and NAD+ rising and two enzymes of polyamine metabolism being switched up. That study used a genetic technique, not 5-Amino-1MQ, so it describes the target rather than this molecule. The Galveston group set out to do the same thing with a drug, and a 2017 Journal of Medicinal Chemistry paper identified quinoliniums as the promising series.
5-Amino-1MQ Benefits
Benefit 1: Less fat gain
What the research describes. The 2018 Biochemical Pharmacology study treated diet-induced obese mice with a potent NNMT inhibitor and reported reduced body weight, reduced white fat mass and smaller fat cells, with total food intake unchanged. The 2024 paper in Diabetes, Obesity and Metabolism dosed the compound once daily for 28 days in diet-induced obese mice and reported dose-dependent limiting of body weight and fat mass gain.
Evidence type. Animal.
What it means. These are the findings most often quoted for 5-Amino-1MQ, and they are mouse findings. Mouse body-composition results frequently do not translate to people, and this compound has not been tested for translation. There is no human trial showing weight loss or fat loss from 5-Amino-1MQ. If losing weight is the actual goal, the best peptides for weight loss sets out the medications with human evidence for it.
5-Amino-1MQ Benefits
Benefit 2: Higher NAD+ inside cells
What the research describes. The 2018 Biochemical Pharmacology paper measured what NNMT inhibitors did inside cultured fat cells: lower 1-methylnicotinamide, the enzyme's product, and higher intracellular NAD+ and SAM.
Evidence type. Cell.
What it means. This is the origin of the "raises NAD+" description. It is a cell-culture measurement and has not been reproduced in human tissue; whether 5-Amino-1MQ changes NAD+ in a person has never been measured. It does not contain NAD+ and does not supply it. If NAD+ itself is the interest, NAD+ injection supplies the molecule directly, and NAD+ benefits covers its own evidence.
5-Amino-1MQ Benefits
Benefit 3: Less fat-building inside fat cells
What the research describes. The same 2018 cell work reported suppressed lipogenesis, the building of new fat within the cell, in cultured fat cells treated with NNMT inhibitors.
Evidence type. Cell.
Strength. A laboratory measurement in a dish. It is a plausible link between the enzyme and the mouse results, and the furthest of these benefits from anything a person would notice.
5-Amino-1MQ Benefits
Benefit 4: Blood sugar and insulin sensitivity
What the research describes. The 2024 Diabetes, Obesity and Metabolism paper reported improved oral glucose tolerance and insulin sensitivity and suppressed hyperinsulinaemia, meaning less excess insulin in the blood, in diet-induced obese mice treated for 28 days.
The human angle. A 2015 study in Diabetologia measured NNMT in fat tissue from 199 people undergoing abdominal surgery and reported it roughly twofold higher in people with type 2 diabetes, with plasma 1-methylnicotinamide tracking tissue NNMT and both correlating with insulin resistance. Exercise and bariatric surgery were associated with lower NNMT expression. Nobody in that study was given 5-Amino-1MQ.
Evidence type. Animal, plus human observation of the target enzyme.
Strength. The best argument for studying 5-Amino-1MQ in people, and still not evidence that it does anything in them. It is not a treatment for diabetes. If you take anything that lowers blood sugar, it belongs on your intake.
5-Amino-1MQ Benefits
Benefit 5: Fatty liver and liver enzymes
What the research describes. The 2024 mouse paper reported less hepatic steatosis (fat in the liver) and less macrophage infiltration on liver histology, and normalisation of the liver enzymes ALT and AST and of ketone bodies, in treated obese mice.
Evidence type. Animal.
What it means. One mouse study. It is also part of the safety picture: in those mice, liver enzymes moved toward normal rather than rising. It is not evidence of any effect on the human liver, and it is not a treatment for liver disease.
5-Amino-1MQ Benefits
Benefit 6: Cholesterol
What the research describes. The 2018 Biochemical Pharmacology study reported lower plasma total cholesterol in treated diet-induced obese mice.
Evidence type. Animal.
Strength. A secondary finding in one mouse study. Nothing in people.
5-Amino-1MQ Benefits
Benefit 7: Working alongside a diet change
What the research describes. A 2022 study in Scientific Reports combined the inhibitor with a switch from a high-fat Western diet to a low-fat diet in diet-induced obese mice, and reported that the combination normalised adiposity and weight to those of lean animals faster than the diet switch alone, with a distinct gut microbiome pattern in treated mice.
Evidence type. Animal.
What it means. A small mechanistic study, and it says nothing about a person who has not changed their diet. It is one reason prescribers describe 5-Amino-1MQ as an adjunct to diet and activity, not a replacement.
5-Amino-1MQ Benefits
Benefit 8: Aged muscle and strength
What the research describes. A 2019 paper in Biochemical Pharmacology from the Galveston group described NNMT as overexpressed in aged skeletal muscle and linked it to impaired NAD+ salvage and senescence of muscle stem cells. In 24-month-old mice given an acute muscle injury, treatment was reported to raise muscle stem cell activity and support larger regenerated muscle fibres. A 2024 paper in Scientific Reports treated mice from 22 to 24 months of age with the inhibitor, intensive exercise, or both, and reported greater grip strength in treated sedentary mice than in sedentary controls, with an additive effect when combined with exercise.
Evidence type. Animal.
What it means. The mice were chosen because they recapitulate features of human sarcopenia, age-related muscle loss. That is a rationale for studying it in older people; it is not a study in people, and 5-Amino-1MQ is not a treatment for muscle loss.
5-Amino-1MQ Benefits
Benefit 9: Appetite, and what it is not
What the research describes. The 2018 mouse work reported total food intake unchanged in treated animals.
What it means. The mechanism described is inside fat cells, not in the appetite centers of the brain. 5-Amino-1MQ is not a GLP-1 medication and should not be expected to reduce hunger. That is a difference in mechanism, and it is why some physicians prescribe it alongside a GLP-1 rather than instead of one; no human study has tested that combination. See what peptides can you take with tirzepatide.

What the evidence does not establish
- That 5-Amino-1MQ does anything in a human being. Mouse body-composition findings frequently do not translate, and this compound has not been tested for translation.
- An effect size. No human trial has measured one.
- A timeline in humans. The 28-day mouse study is the closest thing to a duration reference.
- That it raises NAD+ in people.
- Long-term safety in humans. There is no human safety database of any size.
- An interaction profile. NNMT also methylates foreign compounds, which is a theoretical reason for care, and nothing more is known.
- Efficacy for any condition as an FDA-approved treatment. It is not approved for anything.
Physicians have prescribed compounded 5-Amino-1MQ for several years, and that experience is real, but it is uncontrolled. The honest framing is "described in mouse and cell research and used clinically," not "shown to work in people." Anyone describing human results is describing something that does not exist.
Where it stands with the FDA
5-Amino-1MQ is not an FDA-approved drug, is not a component of one, and has no USP monograph. It is not on the 503A bulks list, and it does not appear in Category 1, 2 or 3 of the FDA's nominated-substances list as updated 14 May 2026. The FDA has never published a safety-risk finding about it, and it was not among the substances the Pharmacy Compounding Advisory Committee reviewed in July 2026. It has simply not been through the FDA's evaluation process. It remains a compounded, prescription-only medication, and compounded medications are not FDA approved. See are peptides FDA approved.

How it compares with MOTS-c and SLU-PP-332
These are the three metabolic options people most often compare, and none has a randomised human trial.
| 5-Amino-1MQ | MOTS-c | SLU-PP-332 | |
|---|---|---|---|
| A peptide? | No | Yes | No |
| Target | The enzyme NNMT | AMPK, via the folate cycle | ERRα, ERRβ, ERRγ |
| Main tissue in the research | Fat tissue, also liver and aged muscle | Skeletal muscle | Skeletal muscle first |
| Human data | None; target enzyme measured in human fat | Observational only | None |
| Price at Pepti | $219/mo | $259/mo | $229/mo |
No study has compared any two of them. The useful difference is where each acts, not which is "strongest". The full side-by-side is in MOTS-c vs SLU-PP-332 vs 5-Amino-1MQ, and the MOTS-c evidence is in MOTS-c benefits.

Side effects, in brief
| What is reported | Where it comes from | What to do |
|---|---|---|
| Injection-site redness, mild soreness, occasional bruising | What prescribers report | Rotate sites |
| No observable adverse effects at the doses used | The 2018 mouse work, not a toxicology study | Not a human safety finding |
| A reaction that is severe, spreading, or involves difficulty breathing | Rare | Stop and get emergency care, then tell your physician |
There is no human safety database and no established interaction list, which is why the intake asks for every medication and supplement you take. 5-Amino-1MQ is not used in pregnancy or breastfeeding. Tested athletes should check the current prohibited list for their sport. The full list is in 5-Amino-1MQ side effects.

Common questions
5-Amino-1MQ Benefits, answered.
In diet-induced obese mice, 5-Amino-1MQ was reported to limit weight and fat gain, improve glucose tolerance and insulin sensitivity, and reduce fatty liver, with food intake unchanged. In cultured fat cells it raised NAD+ and SAM. In aged mice it was reported to support muscle repair and grip strength. None of this has been tested in people.
In laboratory work it blocks the enzyme NNMT, which uses up an NAD+ precursor and the methyl donor SAM. Blocking it is proposed to keep both in the cell and shift fat cells toward using energy. That is described in cells and mice only.
There is no human trial of 5-Amino-1MQ for weight loss or anything else. In obese mice it limited weight and fat mass gain. If weight loss is the goal, semaglutide and tirzepatide are the medications with human trial evidence.
In cultured fat cells, NNMT inhibitors were reported to raise intracellular NAD+ and SAM. Whether 5-Amino-1MQ changes NAD+ in a person has not been measured. It does not contain NAD+.
Not as described in the research. The 2018 mouse study reported food intake unchanged in treated animals, and it is not a GLP-1 medication.
No study has tested that in people. In aged mice, it was reported to support muscle stem cell activity after injury and greater grip strength, with an additive effect alongside exercise. That is a rationale for research, not evidence in people.
No. There are no human trials of any design. The only human data concerns the target enzyme: a 2015 Diabetologia study reported NNMT roughly twofold higher in the fat tissue of people with type 2 diabetes.
There is no human timeline, because there is no human study. The most relevant mouse study ran 28 days, which is about the span of one vial, but a mouse timeline is not a human one.
No. It is a small synthetic molecule, 5-amino-1-methylquinolinium, with no amino acids in it. It is grouped with peptides because of how it is prescribed and injected.
No. It is not an approved drug or a component of one, and compounded medications are not FDA approved. The FDA has not evaluated it for compounding in either direction and has never published a safety-risk finding about it.
At Pepti, the injectable vial is $219 a month, all-in, covering medication, physician review, refill management and shipping. The FUSION Blend, which contains it, is $279 a month.
No. Bloodwork is not required for any Pepti treatment. If your physician wants labs for your care, they'll ask through your account. The free assessment goes to a physician licensed in your state. A consultation does not guarantee a prescription.
