Melanotan II Dosage: How Pepti Prescribes It, in Units, mL and mg
This page explains the Melanotan II dosage Pepti prescribes, what the units on the syringe mean, a chart that converts units to millilitres and milligrams, why the schedule is weekdays only, how long a vial lasts, what the human studies did and did not measure, the side effects and skin checks that come with it, and where it stands with the FDA. It covers how Pepti's Melanotan II is typically prescribed, not dosing protocols from the internet. Nothing here is a reason to change your own dose.

Treatments mentioned
At Pepti, Melanotan II is prescribed as 0.25 mL, which is 25 units on a U-100 insulin syringe, injected under the skin once daily Monday through Friday. The vial is 5 mL at 2 mg/mL, so each dose carries 0.5 mg of Melanotan II and the vial holds 20 doses, about four weeks. Your physician sets your directions after reviewing your intake, and your exact dose and units are printed on the vial. Melanotan II is a compounded prescription medication; it is not FDA approved, and it is scheduled for review by the FDA's compounding advisory committee before the end of February 2027.

Melanotan II Dosage
The short answer
| Product | Vial | Typical directions | Melanotan II per dose | Doses per vial | Price |
|---|---|---|---|---|---|
| Melanotan II | 5 mL, 2 mg/mL (10 mg per vial) | Inject 0.25 mL (25 units) subcutaneously once daily Monday through Friday | 0.5 mg | 20 | $249/mo |
The price is all-in, covering medication, physician review, refill management and shipping, with no separate membership fee. Pepti is cash-pay and does not bill insurance. Melanotan II is a vial and syringe only; there is no pen. These are the typical printed directions; your own directions are the ones on your vial.
Melanotan II Dosage
Melanotan II dosage in units, mL and mg
Pepti's directions are written in two forms, millilitres and units, because prescriptions are written in millilitres and insulin syringes are marked in units. On a U-100 insulin syringe, 1 unit is 0.01 mL, so 100 units is 1 mL and 25 units is 0.25 mL. A unit is a volume mark on the barrel, not a measure of how strong the medication is. "Melanotan 2 dosage in units" is one of the most searched questions about this peptide, and the answer is that units only mean something alongside the concentration on the label.
Pepti's Melanotan II is supplied at 2 mg/mL, so:
| What you read | Value | Where it comes from |
|---|---|---|
| Dose volume | 0.25 mL | Printed on the vial |
| On a U-100 insulin syringe | 25 units | 0.25 mL x 100 units per mL |
| Concentration | 2 mg/mL | Printed on the vial |
| Melanotan II per dose | 0.5 mg | 2 mg/mL x 0.25 mL |
| Total in the vial | 10 mg in 5 mL | Printed on the vial |
| Doses per vial | 20 | 5 mL divided by 0.25 mL |
That arithmetic is a way to read a label, not a way to set a dose. A vial labelled 10 mg is not a 10 mg dose: it is twenty 0.5 mg doses. If your label and your syringe seem to disagree, do not dose; message your care team. How long does a peptide vial last explains every number on a vial label in more detail.
Melanotan II Dosage
Melanotan II dosage chart: units to mL to mg
Online Melanotan II charts mix milligrams, micrograms and units, and many are written for powder vials mixed at home to a concentration nobody can check. The chart below is the conversion for Pepti's 2 mg/mL vial, drawn with a U-100 insulin syringe. It is math only. It is not a menu of doses, and the only line that applies to you is the one that matches the number printed on your vial.
| Syringe mark (U-100) | Volume | Melanotan II from a 2 mg/mL vial |
|---|---|---|
| 5 units | 0.05 mL | 0.1 mg |
| 10 units | 0.10 mL | 0.2 mg |
| 15 units | 0.15 mL | 0.3 mg |
| 20 units | 0.20 mL | 0.4 mg |
| 25 units (Pepti's typical directions) | 0.25 mL | 0.5 mg |
| 30 units | 0.30 mL | 0.6 mg |
| 50 units | 0.50 mL | 1 mg |
| 100 units | 1 mL | 2 mg |
The rule behind every row: units divided by 100 gives millilitres, and millilitres multiplied by 2 gives milligrams. A chart from another source, written for a different concentration, gives different milligram figures for the same units. That is the main reason not to dose from a chart, including this one, and the reason the FDA's case reports of toxicity matter: they involved self-sourced product at doses far above any reviewed direction.

How long a Melanotan II vial lasts
At Pepti's typical directions, one 10 mg vial lasts four weeks:
| Step | Value |
|---|---|
| Dose | 25 units, 0.25 mL, 0.5 mg |
| Doses per week | 5 (Monday through Friday) |
| Doses per vial | 20 (5 mL divided by 0.25 mL) |
| Melanotan II used | 20 x 0.5 mg = 10 mg, the whole vial |
| Time per vial | 20 doses divided by 5 per week = 4 weeks |
Four weeks is why refills run every 28 days, one vial per fill. Vials are multi-dose solutions shipped ready to use, refrigerated with ice packs, with sterile syringes and alcohol prep pads. Nothing is mixed at home. Keep the vial in the fridge, and follow the beyond-use date printed on the label, which runs from first puncture. How to store peptides covers storage.
If a vial runs short before the refill, message your care team rather than adjusting anything yourself. If you miss a dose, follow your directions and do not take two doses to make up for one.

When and how Melanotan II is taken
Monday through Friday
Pepti's directions are five days a week with the weekend off. The 1996 pilot study, the first to measure pigmentation in people, dosed on alternating weekdays over two weeks. The weekday pattern gives a predictable break from the side effects each week, and it makes the vial arithmetic clean: 20 doses at five a week is exactly four weeks. Weekends off are part of the schedule, not missed doses.
Time of day
The typical directions say once daily and do not set a time. If your physician directs a particular time of day, it is printed on your label.
Under the skin, and where
Subcutaneous means into the fat layer, not the muscle. Abdomen and thigh are the usual sites, rotated so the same spot is not used repeatedly. Because the melanocortin signal is systemic, where you inject does not decide where pigment changes. Technique is in how to inject peptides safely.

What the human studies used, and why that is not your dose
Melanotan II has a small human literature, all from one research group in the 1990s:
| Study | Who | What was done and reported |
|---|---|---|
| Pilot phase I study, Life Sciences, 1996 | 3 healthy men | Subcutaneous Melanotan II or saline on alternating weekdays for two weeks, with dose escalation; increased pigmentation measured in 2 of 3 after five low doses |
| Crossover study, Journal of Urology, 1998 | 10 men with psychogenic erectile dysfunction | Single doses against placebo; erections in 8 of 10 |
| Crossover study, Urology, 2000 | 10 men with organic erectile dysfunction | Erections after 12 of 19 active injections vs 1 of 21 placebo |
| Pooled review, International Journal of Impotence Research, 2000 | 20 men | Erections in 17 of 20; increased desire after 68% of active doses vs 19% of placebo |
The honest reading: the pigmentation evidence is one three-person, single-blind pilot that showed an effect but was not designed to measure how much, how evenly, or how long it lasts. There are no large randomised trials of Melanotan II and no controlled trials of repeated dosing over weeks or months, which is how it is prescribed. Nothing characterises how dark or how even a result will be for a given person, how long pigmentation persists after stopping, or the long-term safety of repeated courses. Response varies with skin type, baseline pigmentation and sun exposure. That is why your directions come from a physician who has reviewed your intake, not from a study table or a forum.
Melanotan II is not a sunscreen and does not block ultraviolet light; the research does not describe it preventing sunburn or skin cancer. Nothing about it is a reason to use sunbeds or skip sunscreen.

Skin checks come with the prescription
Melanotan II darkens existing moles and freckles. Case reports describe new moles appearing and existing ones changing during use, and a small number describe melanoma diagnosed during or shortly after a course. A 2017 review in the International Journal of Dermatology concluded that a causal link has not been established but that the reports justify caution and skin surveillance.
| What to do | Why |
|---|---|
| Raise any history of melanoma or atypical moles at intake | It is a stop, not a caveat |
| Have existing pigmented lesions examined before starting | A darker mole is expected; you need a baseline to spot one that changes shape, border or texture |
| Check your skin regularly while on it | The single most important practical point |
| Report any new pigmented spot or changing mole promptly | Message your care team and have it looked at |

What changes the directions
Your physician sets your directions after reviewing your intake. Things that can lead to different directions, or to Melanotan II not being prescribed, include:
| What can change | How it shows up |
|---|---|
| Skin history | A personal history of melanoma or atypical moles is a stop |
| Very fair skin that burns rather than tans | Prescribers describe a wide range of response, and the balance may not be there |
| Your other medications | There is no established interaction list, so the intake asks for everything, including anything for erectile dysfunction or blood pressure |
| How you respond | Marked nausea, or any skin change, goes to your physician, who may revise your directions |
| The goal | If the goal is sexual function rather than pigmentation, PT-141 was developed for that; see PT-141 vs Melanotan II |
When directions change, the new numbers are printed on the new label. Never carry a number over from a previous fill. Bloodwork is not required for any Pepti treatment. If your physician wants labs for your care, they'll ask through your account.

Side effects, in summary
The full reference is what is Melanotan II. The human studies recorded these, and described them as dose-related and most pronounced at the start:
| What is reported | When | What to do |
|---|---|---|
| Nausea | The most commonly reported effect, usually within an hour or two of a dose | Tell your care team if it is a problem |
| Facial flushing and warmth | Shortly after a dose, settling the same day | Usually short-lived |
| Stretching and yawning | Often before other effects; transient and dose-related | Expected, worth knowing in advance |
| Reduced appetite | Recorded on side-effect questionnaires | It is a side effect, not a weight treatment |
| Sleepiness | Reported at the top of the study dose escalation | Report it |
| Spontaneous erections in men | Intermittently for one to five hours after a dose | Expected; an erection lasting more than four hours needs emergency care |
| Darker moles and freckles; new or changing moles | Over weeks | Report any change promptly |
| An erection lasting more than four hours, a severe headache with visual change or confusion, or a racing heart with agitation and muscle pain | Not routine | Call 911 or go to an emergency department |
For any reaction that is severe, spreading, or involves difficulty breathing, stop and contact your physician, and call 911 if it is severe. Melanotan II is not named on the 2026 WADA Prohibited List; tested athletes should check their own sport's current rules. Pregnancy and breastfeeding are exclusions, because safety data is absent.

Where Melanotan II stands with the FDA
Most of what is online about Melanotan II's status is out of date. As the FDA's own pages stood when Pepti's references were verified:
| Date | Status |
|---|---|
| 2023 | Placed in Category 2 of the FDA's 503A bulk drug substances list, the category for substances flagged for significant safety concerns |
| 22 April 2026 | The FDA's safety-risk page lists Melanotan II among substances previously in Category 2 and keeps its original note on file: a possible immunogenicity risk from aggregation or peptide impurities, and published case reports of melanoma, posterior reversible encephalopathy syndrome, sympathomimetic toxidrome and priapism |
| 14 May 2026 | No longer in Category 2 on the FDA's updated list |
| July 2026 | Not among the peptides discussed at the Pharmacy Compounding Advisory Committee meeting |
| Before the end of February 2027 | The committee is scheduled to discuss Melanotan II for possible inclusion on the 503A bulks list, alongside LL-37, GHK-Cu, Dihexa and PEG-MGF |
So the accurate description today is: no longer flagged in Category 2, scheduled for formal advisory committee review by February 2027, and awaiting that review. It remains legal to prescribe and dispense with a valid prescription. It is not an FDA-approved drug, and compounded medications are not FDA approved. The committee's vote will be advisory; the FDA issues its own determination afterwards. See are peptides FDA approved.

Where to get Melanotan II prescribed
Melanotan II cannot be bought legitimately without a prescription. Sites selling it "for research use only" are not accountable for what is in the vial: identity, purity, sterility and concentration are untested, so even the concentration your units chart depends on is unknown. The prescription route:
- Complete a free online intake covering your history, medications, skin history and what you want help with.
- A physician licensed in your state reviews it, usually within 24 hours.
- If it is appropriate, a PCAB-accredited, FDA-registered US 503A compounding pharmacy prepares it to that prescription. Lot test results are published at /quality/lab-results.
- It ships refrigerated with your directions printed on the vial, with sterile syringes and alcohol prep pads.
- Your physician stays reachable for questions and dose changes.

Common questions
Melanotan II Dosage, answered.
At Pepti, Melanotan II is typically prescribed as 0.25 mL, which is 25 units on a U-100 insulin syringe, injected subcutaneously once daily Monday through Friday. From the 2 mg/mL vial that is 0.5 mg per dose. Your physician sets your directions after reviewing your intake, and your exact dose is printed on the vial.
The number printed on your vial. Pepti's typical directions are 25 units, which is 0.25 mL on a U-100 insulin syringe, where 1 unit is 0.01 mL.
From a 2 mg/mL vial, 0.5 mg. 10 units is 0.2 mg and 50 units is 1 mg. Those figures only hold at 2 mg/mL.
On Pepti's typical directions, once daily Monday through Friday, with weekends off. Never take two doses in one day, and do not take two doses to make up for a missed one.
About four weeks. A 10 mg vial at 25 units (0.5 mg) five days a week is 20 injections, which uses the whole vial in four weeks, matching the 28-day refill.
Pepti's typical directions are a single, steady weekday dose; there is no separate loading phase on the label. Take the dose printed on your vial. If you have questions about your dose, message your care team.
There is no trial that answers that. The 1996 pilot measured increased pigmentation in two of three men a week after five low doses, but it was not designed to say how much, how evenly or for whom. Response varies with skin type and sun exposure, and the first week is mostly about side effects.
The pilot measured pigmentation in areas the volunteers had not been exposing, so the pathway does not require ultraviolet light. Nothing about this medication is a reason to use sunbeds or skip sunscreen; it does not prevent sunburn.
No. Take the dose printed on your vial, on the schedule printed on your vial. The FDA's case reports of toxicity and priapism involved self-sourced product at doses far above any reviewed direction. If you have questions about your dose, message your care team.
No. It was placed in Category 2 in 2023 and is no longer in that category on the FDA's list updated 14 May 2026. The FDA's advisory committee is scheduled to discuss it before the end of February 2027. It remains legal to prescribe with a valid prescription, and it is not FDA approved.
No. Pepti vials are multi-dose solutions shipped ready to use, with sterile syringes and alcohol prep pads. Nothing is reconstituted at home.
